Microbiology Calculations?
We did a microbiology lab and I'm just wondering how to do some of the calculations
We removed 200ul from the untreated water sample and added it to the C10^-1 test tube. 200ul was removed from C10^-1 and added to the C10^-2 test tube. 200ul was removed from C10^-2 and added to the C10^-3 test tube. 200ul was removed from C10^-3 and added to C10^-4. The same was repeated with the treated water.
C10^-1, C10^-2, C10^-3 had too many colonies of bacteria to count, C10^-4 had 69 colonies.
T10^-1, T10^-2, T10^-3 had too many colonies of bacteria to count, T10^-4 had 63 colonies.
Calculate the cfu/mL for the control plate and the treatment plate.
What is missing from the cfu/ml calculations that would make the results of the calculations more accurate?
How many bacteria were present in the entire contaminated water sample?

We also added 900ul of saline to each of the tubes before we added the 200ul of water
First, what is the difference between the C and the T tubes. I will assume it was untreated versus treated.
This information is missing: how much diluent medium (PBS or broth) was in the test tubes? One needs to know that because if you add 200 ul to 1800 ul, you will have a 1:10 dilution, but if it was 3800 ul, it would be a 1:20 dilution.
Then, you did not provide how much of each tube you plated out on the plates.
Without that information, I cannot calculate how many bacteria were present in the original sample. But I can tell you that the treatment did not make a significant impact, since the numbers were very similar.
